High-throughput sample preparation for PCR and molecular diagnostics

Molecular diagnostic methods such as polymerase chain reaction (PCR) rely on reproducible sample preparation to generate reliable analytical results. In modern high-throughput systems, large numbers of samples are processed automatically, for example in infectious and respiratory disease diagnostics, transplant monitoring, and oncology.

During sample preparation, nucleic acids such as DNA and RNA are isolated from patient samples. Different reagents and buffer solutions are precisely dosed in volumes ranging from 10 to 5000 µl. At the same time, washing and cleaning steps are performed at high flow rates to support continuous operation with high sample throughput.
 

What requirements does high-throughput sample preparation for PCR applications impose?

Processing large numbers of samples requires dosing technology that combines precision, dynamics, and long-term stability. In addition to dosing small volumes, rinsing cycles with high flow rates are part of the process.

How is dosing implemented in high-throughput systems?

The system uses the mzr-4622 micro annular gear pump with an application-specific drive for OEM integration into automated diagnostic instruments.

The pump performs both precise dosing of small reagent volumes and delivery of larger rinsing volumes. This allows sample preparation and fluidic cleaning to be implemented using the same pump technology.

For abrasive and crystallizing media, ceramic and stainless steel 316L are used as wetted materials. The selected material combinations are designed for long operating times and achieve service lives of up to 3000 operating hours.

Depending on the required sample throughput, different numbers of pumps are integrated into the diagnostic instrument. Due to the compact design, up to 30 micro annular gear pumps or more can be arranged in a space-saving way within one dosing unit.
